Absolutamente! E pelos vistos não há maneira de as pessoas se convencerem dos riscos (e crueldade) da situação. A apresentação foi num congresso da sociedade alemã de veterinária e pelo que eu percebi não importa quanto os donos são avisados, ficam sempre chocados quandos os problemas acontecem.

Eu já não tinha grande atração por cães e gatos de raça e sabia de problemas físicos com o inbreading mas o ano passado assisti a uma apresentação sobre os problemas associados a algumas raças e que só têm aumentado que ainda me deixou mais chocada. É pura crueldade.

These old photos are always so cute. To think that someone over 100 years ago considered it worthwhile to take a photo, an act much more cumbersome and expensive than today, to document a kitten doing something adorable shows that some things never change.

In germ cells from infertile men we found aberrant DNA methylation in genes specifically expressed during spermatogenesis. Moreover, we detected hypomethylation of some evolutionary young transposable element families 🤯 8/9

Then we turned to male infertility, specifically men who produce very few sperm: cryptozoospermia. There were over 100 differentially methylated regions per cell type. Again, imprinting control region methylation remained stable (we made sure our samples were extremely pure) 7/9

Methylation remained stable at imprinting genes. Regions that were affected were enriched in SINE elements but depleted in LINEs 🧐 Also exciting was that spermatid-hypomethylated promoters were associated with spermatid-specific gene expression. How cool is that? 6/9

Lara Siebert-Kuss and Verena Dietrich started analysing the data and found the dip in spermatocyte methylation but it did not seem to be solely a side product of maintenance delay because the remethylation process established a distinct spermatid-specific methylation pattern. 5/9

Sara Di Persio from the CeRA and Jahnavi Bhaskaran from the Vaquerizas lab (now MRC London) were successful and we managed to get enough material for Lara Siebert-Kuss to isolate DNA and perform high quality whole methylome sequencing on pure spermatogonia, primary spermatocytes, and spermatids. 4/9

However, this process was never studied in human, likely due to how challenging isolating pure germ cell types is compared to rodents. We teamed up with our then neighbours from the MPI in Münster to establish a reliable protocol for male germ cell sorting from tiny biopsies 3/9

#DNAmethylation is considered a very stable #epigenetic mark and the methylome of the male germline was long thought to remain stable after it's initial setup in early development. A couple of rodent studies questioned this and showed a decrease in methylation around meiosis. 2/9
