In germ cells from infertile men we found aberrant DNA methylation in genes specifically expressed during spermatogenesis. Moreover, we detected hypomethylation of some evolutionary young transposable element families 🤯 8/9

Then we turned to male infertility, specifically men who produce very few sperm: cryptozoospermia. There were over 100 differentially methylated regions per cell type. Again, imprinting control region methylation remained stable (we made sure our samples were extremely pure) 7/9

Methylation remained stable at imprinting genes. Regions that were affected were enriched in SINE elements but depleted in LINEs 🧐 Also exciting was that spermatid-hypomethylated promoters were associated with spermatid-specific gene expression. How cool is that? 6/9

Lara Siebert-Kuss and Verena Dietrich started analysing the data and found the dip in spermatocyte methylation but it did not seem to be solely a side product of maintenance delay because the remethylation process established a distinct spermatid-specific methylation pattern. 5/9

Sara Di Persio from the CeRA and Jahnavi Bhaskaran from the Vaquerizas lab (now MRC London) were successful and we managed to get enough material for Lara Siebert-Kuss to isolate DNA and perform high quality whole methylome sequencing on pure spermatogonia, primary spermatocytes, and spermatids. 4/9

#DNAmethylation is considered a very stable #epigenetic mark and the methylome of the male germline was long thought to remain stable after it's initial setup in early development. A couple of rodent studies questioned this and showed a decrease in methylation around meiosis. 2/9

Aqui na Alemanha houve um belo documentário no canal Arte e pelo menos uma reportagem na ZDF (tv pública). Estou agora a ouvir um programa de rádio pública sobre a importância da música na revolução. Não sei se houve alguma primeira página, mas houve artigos nos principais jornais.
